Annexin V属于胞内膜联蛋白家族，以钙依赖的方式与膜磷脂结合。这种染料对存在于质膜内小叶中的磷脂酰丝氨酸（PS）有很高的亲和力。在早期细胞凋亡过程中，PS从胞膜的内叶向外叶易位，暴露于胞外环境中。Annexin V是早期细胞凋亡的特征性标志物，可检测PS向胞外环境的易位。Annexin V与7-AAD（产品号 #75001）或碘化丙啶（PI，产品号 #75002）等活性染料一起使用。由于PS易位的过程发生在细胞膜完整性丧失之前，当细胞经历凋亡并走向坏死时，细胞膜受损，导致活性染料进入细胞。因此，发生早期凋亡的细胞Annexin V染色阳性，活性染料染色阴性，而细胞凋亡性死亡或坏死的特征是Annexin V和活性染料染色双阳性。
