生长分化因子-15 (GDF-15),又称巨噬细胞抑制因子-1 (MIC-1),是转化生长因子β (TGF-β)超家族的成员。在人体细胞组织中,生理性的GDF-15表达水平较低或不存在,但GDF-15水平升高与细胞应激、炎症、代谢疾病、癌症和其他疾病有关(Emmerson等;Wischhusen等)。GDF-15通过GFRAL (GDNF Family Receptor Alpha Like)及其共受体RET进行信号转导,导致RET的磷酸化,并激活ERK和AKT信号通路(Emmerson 等)。人重组GDF-15已用于小鼠研究(Kim等)。该产品经检测TGF-β活性为阴性,TGF-β是商业来源GDF-15的常见污染物(Olsen等)。本产品不含动物成分(ACF)。

(A) Human Recombinant GDF-15, ACF was tested by its ability to activate SMAD2/3 in HEK293T cells using a luciferase reporter assay. Firefly luciferase activity is normalized to the control Renilla luciferase activity. Increasing concentration of Human Recombinant GDF-15 did not result in increasing SMAD2/3 activity, indicating it is free of contamination with TGF-β or related growth factors.
(B) 7 μg of Human Recombinant GDF-15, ACF was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Human Recombinant GDF-15, ACF has a predicted molecular mass of 25 kDa (dimer).
