白介素21 （IL-21）是一种多效性细胞因子，由四个α-螺旋束组成，主要由自然杀伤T细胞（NKT）、滤泡辅助性T细胞（Tfh）以及Th17细胞产生（Spolski & Leonard 2008）。IL-21 的信号传导依赖于 IL-21 受体（IL-21R）与编码于 IL2RG 的通用细胞因子受体γ链形成的异源二聚体(Parrish-Novak et al.; Ozaki K et al. 2000)，并通过JAK/STAT、MAPK和PI3K信号通路发挥作用（Spolski & Leonard 2014）。IL-21在调节免疫球蛋白的产生和促炎Th17细胞群的分化中起关键作用(Ozaki et al. 2002; Nurieva et al.)。此外，IL-21能有效维持CD8+ T细胞效应功能，并在慢性病毒感染期间为提供CD4+ T细胞提供协助机制（Elsaesser et al.）。IL-21信号也被发现对非肥胖型糖尿病（NOD）小鼠1型糖尿病的发展至关重要（Sutherland et al.），并通过调节性B细胞控制T细胞自身免疫（Yoshizaki et al.）。

(A) The biological activity of Human Recombinant IL-21 was tested by its ability to inhibit the proliferation of Mino cells. Cell proliferation was measured using a fluorometric assay method. The EC50 is defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum. The EC50 in the example above is less than 0.5 ng/mL. (B) 2 μg of Human Recombinant IL-21 was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Human Recombinant IL-21 has a predicted molecular mass of 15.4 kDa.
