血小板源性生长因子（PDGF）是一种二聚体糖蛋白，由两条二硫键稳定的多肽链A和B组成，可以形成异源二聚体（PDGF- AB）或同源二聚体（PDGF- AA和PDGF- BB）(Fretto et al.; Westermark & Heldin)。PDGF通过受体酪氨酸激酶PDGFRα和PDGFRβ进行信号转导。研究表明，PDGF诱导的细胞迁移涉及多个信号通路，包括MEK/ERK、EGFR、Src和PI3K/AKT通路（Kim et al.）。PDGF对间充质来源细胞（如成纤维细胞、神经胶质细胞和血管平滑肌细胞）有强烈促有丝分裂作用。PDGF被认为在动脉粥样硬化、肾小球肾炎、癌症的发病机制中发挥重要作用，并参与大鼠主动脉组织中血管平滑肌细胞的收缩调节(Fretto et al.; Sachinidis et al.)。PDGF-BB由成骨细胞分泌，诱导间充质干细胞迁移和血管生成。也有研究表明，在骨建模和重塑过程中，PDGF-BB由前破骨细胞分泌，以诱导血管生成，从而促进正常的成骨（Xie et al.）。

(A) The biological activity of Human Recombinant PDGF-BB was tested by its ability to promote the proliferation of NR6R-3T3 cells. Cell proliferation was measured using a fluorometric assay method. The EC50 is defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum. The EC50 in the example above is 2.12 ng/mL. (B) 1 μg of Human Recombinant PDGF-BB was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Human Recombinant PDGF-BB is a homodimer of 12.4 kDa subunits with a predicted total molecular mass of 24.9 kDa.
