R-Spondin-1 (RSPO1)是R-Spondin(RSPO)蛋白家族的原型成员,属于血小板反应蛋白1型重复序列(TSR-1)的蛋白超家族的亚家族(Chen et al. Mol Biol Rep, 2002; Kazanskaya et al. Dev Cell, 2004; Kim et al. Cell Cycle, 2006)。虽然RSPO家族成员本身不能启动信号通路,但却是WNT信号通路的强效增强因子(Cruciat & Niehrs. Cold Spring Harb Perspect Biol, 2013; de Lau et al. Genome Biol, 2012; Kazanskaya et al. (A) The biological activity of Human Recombinant R-Spondin-1 (E. coli-expressed), ACF was tested by its ability to induce luciferase activity in HEK239T cells transfected with the TOP-FLASH reporter. Luciferase activity was measured after cells were treated with increasing concentration of R-spondin-1 (diluted in DMEM with 0.5% of fetal calf serum), in the presence of Wnt-conditioned medium (1 in 8 dilution). The EC50 is defined as the effective concentration of the growth factor at which Wnt-ß catenin activity is at 50% of maximum. The EC50 in the above example is 5.8 nM (75.1 ng/mL). (B) 7 µg of Human Recombinant R-Spondin-1 (E. coli-expressed), ACF was resolved with SDS-PAGE under reducing (+) conditions and non-reducing (-) conditions and visualized by Coomassie Blue staining. Human Recombinant R-Spondin-1 has a predicted molecular mass of 13 kDa in reducing conditions and 16 kDa in non-reducing conditions.
