利用骨硬化蛋白调节骨重塑的动态平衡，可能通过Wnt/β-catenin信号级联，在体内和体外抑制骨形成(Ellies等；Lin等)。Sclerostin是cerberus/DAN糖蛋白家族的成员，其成员具有一个共享C端半胱氨酸结样（CTCK）结构域。这种破骨细胞来源的BMP拮抗剂在在骨和软骨中表达高度，同时也在肾脏和肝脏组织中表达（Weivoda等），它对BMP6和BMP7具有高亲和力，并对BMP2和BMP4具有较低亲和力（Kusu等人）。SOST基因突变与硬化症（Brunkow等）、van Buchem病（Staehling-Hampton等）和以骨密度增加为特征的骨发育不良疾病有关。该蛋白产物在多肽链的羧基端带有一个His标签。为确保各种应用中的一致性与可重复性，STEMCELL公司的硬化蛋白经过冻干，纯度≥87%，内毒素水平被验证为≤1.0 EU/μg蛋白。

(A) The biological activity of Human Recombinant Sclerostin was tested by its ability to inhibit Wnt3a-induced alkaline phosphatase production by C3H10T1/2 cells. The EC50 is defined as the effective concentration of the cytokine at which inhibition of enzyme production is at 50% of maximum. The EC50 in the above example is ≤ 1 μg/mL. (B) Human Recombinant Sclerostin was resolved with SDS-PAGE under reducing (+) conditions and visualized by Coomassie Blue staining. Human Recombinant Sclerostin has a predicted molecular mass of 22.5 kDa but an apparent molecular mass of 29.9 kDa due to glycosylation.
