总览

Annexin V凋亡检测试剂盒包括Annexin V（产品号 #100-0329或产品号 #100-0331或产品号 #100-0333），Annexin V结合缓冲液（产品号 #100-0334），和7-AAD（产品号 #75001.1）。该试剂盒用于联合检测早期细胞凋亡（使用Annexin V）和晚期细胞凋亡或坏死（同时使用Annexin V和7-AAD）。Annexin V属于胞内膜联蛋白家族，以钙依赖的方式与膜磷脂结合。这种染料对存在于质膜内小叶中的磷脂酰丝氨酸（PS）有很高的亲和力。在早期细胞凋亡过程中，PS从胞膜的内叶向外叶易位，暴露于胞外环境中，并被Annexin V检测到。磷脂酰丝氨酸易位过程发生在细胞膜完整性丧失之前。当细胞经历凋亡并走向坏死时，细胞膜受损，导致7-AAD活性染料进入细胞。因此，发生早期凋亡的细胞Annexin V染色阳性，活性染料染色阴性，而细胞凋亡性死亡或坏死的特征是Annexin V和7-AAD染色双阳性。
