RPA-T8抗体识别CD8a，CD8a是一种32 - 34 kDa的I型跨膜糖蛋白，是CD8的一个亚基。CD8是一种二硫键结合的二聚体，可以是CD8a （α）和CD8b （β）亚基的异二聚体（即αβ）或同二聚体（αα）。CD8通过结合抗原呈递细胞呈递的MHC I类分子，在T细胞活化过程中作为T细胞受体（TCR）的共受体。其功能是加强TCR与MHC I-抗原复合物之间的联系，并通过其胞内结构域与细胞质酪氨酸激酶（如Lck）的相互作用，放大从TCR到细胞质的信号。CD8a链与MHC I类分子的α3结构域结合。CD8在大多数胸腺细胞、一部分成熟的外周血T细胞（细胞毒性T细胞）、和一部分β T细胞上表达，在NK细胞上也有较低水平的表达（主要表达CD8a同二聚体）。
