总览

L243抗体可特异性识别人HLA-DR，HLA-DR是主要组织相容性复合物（MHC） II类受体，属于免疫球蛋白超级家族。HLA-DR是一种异源二聚体跨膜糖蛋白，由一个36 kDa的α亚基与一个27 kDa的β亚基以非共价结合而成，表达于抗原递呈细胞（如B细胞、活化T细胞、单核细胞、巨噬细胞和树突状细胞）、活化NK细胞和祖细胞的表面。HLA-DR与CD3/T细胞受体（TCR）复合物和CD4分子协同作用，介导抗原递呈细胞将外源抗原水解产生的肽递呈给CD4+ T（辅助）细胞，从而调控针对这些肽的免疫激活或抑制反应。因此，HLA-DR与移植物抗宿主病和几种自身免疫性疾病相关。L243抗体可结合α链上的一个胞外、构象性和非多态性表位，该表位仅在正确折叠的αβ异源二聚体表达，并且结合不依赖于HLA-DR与抗原肽装载。该抗体的结合阻断HLA-DR与TCR的结合，研究发现其对人细胞具有细胞毒性作用。L243抗体不与HLA-DQ或HLA-DP发生交叉反应。
