2B8抗体识别CD117 (c-Kit),CD117 (c-Kit)是一种~145 kDa的c-Kit配体(干细胞因子/钢铁因子)的1型跨膜受体,广泛表达于骨髓造血干细胞,包括多能祖细胞、红系祖细胞、B和T淋巴细胞前体,以及肥大细胞。CD117属于酪氨酸激酶受体家族,具有5个免疫球蛋白样C2型结构域和1个细胞质蛋白激酶结构域。c-Kit配体与CD117结合可诱导二聚化和自磷酸化,从而激活对造血干细胞增殖和分化至关重要的几种细胞内信号通路。CD117信号通过磷酸酶和受体的快速内吞和降解来调控。2B8抗体不会阻断c-Kit配体的结合。CD117的突变与各种类型的肿瘤和骓斑(一种常染色体显性色素沉着异常)有关。
