人免疫球蛋白G（IgG）ELISpot抗体配对试剂盒旨在为希望灵活设置ELISpot检测方案的用户提供便利。该试剂盒用于检测和计数外周血单个核细胞（PBMCs）悬液中分泌人IgG的细胞。ELISpot常用于评估免疫反应，如循环抗原特异性记忆B细胞或抗体分泌细胞（ASCs）的占比和亚型。可溶性IgG在针对病原体的继发免疫反应中起重要作用。ELISpot具有高灵敏度，可在单细胞水平检测分泌的分析物。该检测方法广泛用于传染病和疫苗研究。

该检测方法可定量抗原特异性和/或总IgG分泌细胞。检测基于改良的夹心ELISA法，将细胞加入预包被有人IgG特异性捕获抗体（方案1）或目标抗原（方案2）的ELISpot板中。在整个细胞培养期间，细胞分泌的IgG被捕获，随后洗去细胞。通过加入生物素标记的抗原或检测抗体来检测捕获的IgG，然后加入SA-ALP与抗原或抗体结合。加入显色酶底物5-溴-4-氯-3-吲哚基-1-磷酸（BCIP）/硝基蓝四唑（NBT）后，会形成有色斑点。这些斑点的数量、大小和强度与样本中抗原特异性或总IgG分泌细胞的占比及分泌的分析物量成正比。

注意：该试剂盒包含足量试剂用于4块96孔ELISpot板。ELISpot板（如Millipore Sigma产品号#MSIPS4W10/MAIPSWU10）和BCIP/NBT底物（产品号#200-1016）为使用该试剂盒所必需，需单独购买。检测抗原特异性免疫反应时还需提供目标抗原。

ELISpot analysis of peripheral blood mononuclear cells (PBMCs) isolated from whole blood using EasySep™ Direct Human PBMC Isolation Kit (Catalog #19654) and cultured in RPMI 1640 Medium (Catalog #36750) supplemented with 2 mM L-glutamine (Catalog #07100) and 10% Fetal Bovine Serum (Catalog #100-0180). Cells were pre-activated with Anti-human CD40 mAb (S2C6), activator + Recombinant human IL-4, lyophilized (included in kit) or in the absence of stimuli (negative control) at 37°C, 5% CO2 for 5 days. Cells were then seeded at 2 x 10^5 cells/well on ELISpot plates pre-coated with Anti-human IgE mAb (107), unconjugated and incubated for a further 20 hours. Following incubation, ELISpot plates were processed as per the instructions on the Product Information Sheet. Cells were then seeded at 1.25 x 10^4 cells/well on ELISpot plates pre-coated with Anti-human IgG mAbs (MT91/145), unconjugated and incubated for a further 18 hours. Following incubation, ELISpot plates were processed as per the instructions on the Product Information Sheet for detecting total IgG. Wells were imaged and counted using an ELISpot reader.
