总览

肝细胞生长因子/分散因子(HGF/SF)是一种多效性生长因子,可促进增殖、迁移、存活和分化。HGF结合并促进受体酪氨酸激酶c-MET的二聚化和激活,刺激PI3K/AKT、FAK、JNK和ERK1/2等信号通路(Organ & Tsao. Ther Adv Med Oncol, 2011)。HGF在胚胎发生过程中促进细胞迁移,诱导上皮细胞的运动和散射,调节上皮-间质转化。其为运动神经元的趋化因子,调节感觉、交感、副交感和皮质神经元的发育,以及促进少突胶质细胞的增殖和神经胶质细胞的发育。此外,HGF具有细胞保护作用,调节肝脏生长和再生(Nakamura et al. J Gastroenterol Hepatol, 2011)。

(A) The biological activity of Human Recombinant HGF (E.coli-expressed), ACF was tested by its ability to induce MAP/ERK signaling in transfected HEK293T cells. Cells were treated with a serial dilution of HGF for 6 hours, and the activity was quantified by a luciferase reporter assay. The EC50 is defined as the effective concentration of the growth factor at which MAP/ERK activity is at 50% of maximum. The EC50 in the above example is ≤ 49.6 pM (0.99 ng/mL). (B) 7 μg of Human Recombinant HGF (E.coli-expressed), ACF was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Human Recombinant HGF has a predicted molecular mass of 21 kDa in reducing conditions and 18 kDa in non-reducing conditions.
