人肿瘤坏死因子 (TNF)-α ELISA 试剂盒用于定量检测细胞培养上清液中的人 TNF-α。TNF-α 主要由单核细胞和巨噬细胞产生，参与调节免疫细胞、细胞增殖、分化、凋亡和脂质代谢。TNF-α 的激活由两种不同的细胞表面受体介导：TNFR1（TNF 受体 1）和 TNFR2（TNF 受体 2）。TNF-R1 具有胞质死亡结构域，导致多种蛋白质降解并引发细胞凋亡。TNF-R2 募集 TNF 受体相关因子 2 (TRAF2) 并诱导基因表达。TNF-α 介导多种疾病，包括癌症、类风湿性关节炎和克罗恩病。

该检测基于夹心ELISA方法，将样品加入预先包被有针对人TNF-α特异性捕获抗体的ELISA可拆酶标板。通过添加辣根过氧化物酶 (HRP)偶联的检测抗体来检测捕获的TNF-α。添加显色酶底物3,3',5,5'-四甲基联苯胺 (TMB)后，显色产物的强度与样品中人TNF-α的浓度成正比。

The reportable range (the concentration range in which measurement of the analyte can be done with the highest precision, accuracy, and linearity) of this assay is 31.25 - 2000 pg/mL. The lower limit of detection (LLD) of this assay is 18.29 pg/mL. This is the analyte concentration with absorbance three standard deviations higher than the zero standard. A mid-curve recovery of 73 - 95% was determined by spiking defined amounts of analyte standard into cell culture supernatant samples in repeated experiments. The intra-assay precision of this assay is 3.8% (CV). The Inter-assay precision of this assay is 3.2% (CV).
