总览

血小板生成素 (TPO) 是体内外巨核细胞生成和血小板生成的关键调节因子。TPO 刺激巨核细胞增殖和成熟，并在调节体内循环血小板水平方面发挥重要作用。TPO 还促进造血干细胞和原始多谱系祖细胞的存活、自我更新和扩增。它通常与其他细胞因子（例如干细胞因子 (SCF) 和 Flt3/Flk-2 配体）一起使用，以促进培养的原始造血细胞扩增 (Hitchcock & Kaushansky)。TPO 受体 c-Mpl 在巨核细胞生成的所有阶段均有表达，从造血干细胞和祖细胞到成熟的血小板。

(A) The biological activity of Mouse Recombinant TPO was tested by its ability to promote the proliferation of MO7e cells. Cell proliferation was measured using a fluorometric assay method. The EC50 is defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum. The EC50 in the above example is 0.92 ng/mL. (B) 5 μg of Mouse Recombinant TPO was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Mouse Recombinant TPO has a predicted molecular mass of 35 kDa but as a result of glycosylation has an apparent molecular mass of approximately 75 kDa.
