总览

小鼠肿瘤坏死因子-α (TNF-α)酶联免疫吸附测定试剂盒用于定量检测和测量小鼠血清、血浆和细胞培养上清等生物液体中的TNF-α。TNF-α主要由活化的T细胞和巨噬细胞在炎症或感染反应中产生，但也由B细胞、NK细胞、肥大细胞、中性粒细胞、树突状细胞、小胶质细胞、内皮细胞、平滑肌细胞、心肌细胞和成纤维细胞产生。TNF-α是一种促炎细胞因子，在体外刺激抗肿瘤反应。

该检测基于夹心ELISA方法，将样品添加到酶联免疫吸附试验（ELISA）带材板上，该带材板上预先涂有捕获的细胞因子特异性抗体。捕获的细胞因子通过添加生物素化的检测抗体来检测，然后添加链霉亲和素-辣根过氧化物酶，该酶与生物素化的抗体结合。添加显色酶底物3,3 ‘,5,5 ’四甲基联苯胺（TMB）可得到着色产物，其强度与样品中细胞因子的浓度成正比。细胞因子的浓度是通过与平行分析的细胞因子标准品的连续稀释进行比较来确定的。

• Reportable range: 15.6 - 2000 pg/mL. This is the concentration range in which measurement of the analyte can be done with the highest precision, accuracy, and linearity.
• Sensitivity: The limit of detection of this assay is 5 pg/mL. This is the analyte concentration with absorbance two standard deviations higher than the zero standard.
• Accuracy: The analyte standard of this ELISA was calibrated against NIBSC international standard 88/532.
• Recovery: A mid-curve recovery of 84 - 86% was determined by spiking defined amounts of analyte standard into serum or plasma samples in repeated experiments.
