总览

HIB22抗体识别CD22 (Siglec-2),CD22是一种I型跨膜糖蛋白,表达于B祖细胞和前体细胞的细胞质以及成熟和活化的B细胞和B细胞淋巴瘤的表面,但不表达于浆细胞。CD22作为B细胞受体信号传导的抑制受体,在调节B细胞的激活阈值和控制CD40信号传导中发挥作用。CD22包括两种选择性剪接产生的异构体:CD22α (~130 kDa)和CD22β (~140 kDa)。单体CD22β是最丰富的亚型,但αβ异源二聚体也有表达。CD22是免疫球蛋白超家族中唾液粘附亚群的成员,具有N端V-type Ig结构域和6个C2-type Ig结构域。据报道,HIB22抗体的结合需要两个最 N 端结构域的存在。CD22的细胞质和表面结合形式都能被HIB22抗体识别,并且它的结合不会抑制CD22介导的细胞粘附。
